چکیده
Crop rotation was practiced in ancient times by alternating the use of leguminous and nonleguminous plants. The basis for the benefit derived from this practice was not understood, but it was observed empirically that crops were improved and the condition of the soil was maintained better with rotation than by continual cropping with nonleguminous plants. Rotation was recognized as good husbandry, but it remained for Boussingault to offer an explanation of the benefit in the 1830's. He established one of the earliest agricultural experiment stations and demonstrated in the greenhouse that clover and peas increased their nitrogen content when grown on sand that was virtually free of fixed nitrogen. His field experiments likewise supported the view that leguminous plants could use N. from the atmosphere. The observations of Boussingault touched off heated controversy and Liebig suggested that the observed increases in nitrogen in the leguminous crops arose merely because the large leaved leguminous plants were capable of extracting more ammonia from the atmosphere than were the smaller leaved control plants. Because Liebig could speak with authority in the area of organic chemistry, he apparently felt that this carried with it the privilege of speaking in the area of agricultural chemistry without the need to present experimental evidence in support of his postulates. It is hardly surprising that a French commission, assigned to the task of checking to see whether Liebig or Boussingault were correct, came to the conclusion that their fellow countryman in fact had demonstrated an increase in nitrogen in the leguminous plants.

Electron Microscopy of the Mo-fe Protein from Azotobacter Nitrogenase
Biological N2 fixation, the reduction of N2 to NH 3 , is catalyzed by the enzyme nitrogenase . Fractionation studies of nitrogenases from a variety of organisms have shown that all are composed of two similar proteins, one contains Mo and Fe and is called the Mo-Fe protein and the other contains only Fe and is called the Fe protein . Recently homogeneous preparations of these proteins from Azot...
متن کاملRegulation of nitrogen fixation in Klebsiella pneumoniae: evidence for a role of glutamine synthetase as a regulator of nitrogenase synthesis.
Mutations causing constitutive synthesis of glutamine synthetase (GlnC(-) phenotype) were transferred from Klebsiella aerogenes into Klebsiella pneumoniae by P1-mediated transduction. Such GlnC(-) strains of K. pneumoniae have constitutive levels of glutamine synthetase. Two of three GlnC(-) strains of K. pneumoniae studied, each containing independently isolated mutations that confer the GlnC(...
متن کامل“Biological Nitrogen Fixation” Book Summary
Biological nitrogen fixation is a very valuable alternative to nitrogen fertilizer. This process will be discussed in the “Biological Nitrogen Fixation” book. A wide array of free-living and associative nitrogen fixing organisms (diazotrophs) will be covered. The most extensively studied and applied example of biological nitrogen fixation is the symbiotic interaction between nitrogen fixing “rh...
متن کاملAutoradiographic Localization of 13n after Fixation of 13n-labeled Nitrogen Gas by a Heterocyst-forming Blue-green Alga
(13)N, generated by proton bombardment of (13)C powder, is rapidly and easily converted to (13)N-N(2), 0.01 atm pressure, ca. 10 mCi/ml, by automated Dumas combustion. (13)N fixed (as (13)N-N(2)) by algal filaments was localized by an autoradiographic technique which permits track autoradiography with isotopes having short half-lives. Our findings show directly that a minimum of about 25% of th...

Volume 24 completes the presentation of methods applicable to the study of the biochemical and biophysical properties of photosynthetic and nitrogen fixing systems. The first part of this volume is concerned with the description of the physical and biochemical methods applicable to the identification of individual components of the photosynthetic system and the determination of their kinetic pa...
